Understanding Your DNA Paternity Test Results

What Your DNA Paternity Test Results Actually Mean

A DNA paternity test compares genetic material between a child and a potential father to determine biological relationship. The results you receive will show one of three outcomes: paternity is confirmed, paternity is excluded, or the results are inconclusive. Understanding what these results actually mean—and what they don't mean—is the foundation for interpreting your test report.

When a test confirms paternity, it means the tested man shares enough DNA markers with the child to be the biological father. Modern DNA tests examine between 15 and 25 genetic locations called loci. At each location, you inherit one genetic variant from your mother and one from your father. A paternity test looks for this inheritance pattern. If the man's DNA matches the child's at nearly all tested locations in the way a biological father's would, the test confirms paternity. The standard threshold for confirmation is 99.9% or higher probability of paternity.

When a test excludes paternity, it means the tested man cannot be the biological father based on the DNA evidence. This happens when the man's genetic markers don't match the child's in multiple locations where they should match if he were the father. Even one clear mismatch at a single locus can exclude someone, though reputable labs test multiple locations to avoid errors.

Inconclusive results are less common but do occur. This might happen if the DNA sample was degraded or contaminated, if the lab couldn't extract enough usable DNA, or if there are technical issues during testing. In these cases, labs typically recommend retesting with new samples.

Practical takeaway: Before reviewing your results, know what outcome you're looking for and what each result category means. Request a detailed report that explains which specific genetic markers were tested and how the lab reached its conclusion.

How DNA Matching Works in Paternity Testing

DNA paternity testing relies on understanding how inheritance works. Every person carries two copies of most genes—one inherited from each parent. When examining a specific genetic location, a person has two alleles (versions of that gene). A child's alleles at any location must come from one parent's pair and one allele from the other parent's pair. This biological rule is absolute and forms the basis of paternity testing.

Consider a simple example. At a particular genetic location, suppose a child has alleles labeled A and B. The child received one of these from the mother and one from the father. If the mother's alleles are A and C, then the child's A came from the mother. This means the child's B allele must have come from the biological father. Therefore, the biological father must carry at least one B allele at this location. If the tested man has alleles C and D at this same location, he cannot be the father because he has no B allele to pass on.

Modern paternity tests examine multiple locations to create a statistical picture. The more locations tested, the more confident the result. Testing 16 locations provides extremely high accuracy. Testing 25 locations increases accuracy further. Each additional location tested reduces the possibility of coincidental matching between an unrelated man and child. This is why reputable labs test numerous locations rather than just one or two.

The lab calculates a Cumulative Index of Paternity (CIP), often presented as a percentage. A result of 99.9% paternity means there's a 1 in 1,000 chance that a random man would match as well as the tested man did. Some labs instead report the Combined Paternity Index (CPI), which expresses how many times more likely the results are if the tested man is the father versus if a random man is the father.

Practical takeaway: Look for results that tested at least 16 genetic locations. Ask your lab how they calculated their probability percentage and what that number actually represents—it's not the same as certainty, but rather a statistical likelihood.

Understanding Probability Percentages and What They Mean

The percentage shown on paternity test results confuses many people. A result showing "99.9% probability of paternity" does not mean there's a 0.1% chance the man is not the father in your specific case. Instead, it's a statistical measure comparing two scenarios: the tested man is the father versus a randomly selected man from the general population is the father. Understanding this distinction matters for interpreting what your results actually tell you.

Think of it this way: the percentage reflects how much more likely your results are under one scenario compared to another. A 99.9% result means the genetic evidence is 999 times more consistent with the tested man being the father than with a random man being the father. This is extremely strong evidence, but it's built on comparing the tested man to the general population, not to other specific individuals.

The lab's calculation uses the child's DNA, the mother's DNA (if available), and the tested man's DNA. When the mother's DNA is included in testing, the results become more precise because the lab can determine exactly which alleles the child inherited from the mother. This makes it clearer which alleles must have come from the father. If the mother's DNA isn't tested, the lab must estimate which alleles she likely contributed, which introduces slightly more uncertainty.

Different labs may report results differently. Some use percentages (99.9%), others use index ratios (like 999:1), and some use both. All reputable labs interpret results the same way: a probability of 99% or higher is considered to confirm paternity. Probabilities below 0% (which you'll sometimes see written as negative values) exclude paternity. Results between these ranges are typically inconclusive and may warrant retesting.

It's worth noting that DNA evidence, while extremely reliable, is just one type of evidence. Courts and legal proceedings may consider other factors alongside test results, and different jurisdictions have different standards for what constitutes legal proof of paternity.

Practical takeaway: Request documentation showing how the lab calculated the percentage. Ask whether the mother's DNA was tested, since this affects the precision of results. Remember that 99.9% is not the same as absolute certainty, but it represents extremely strong scientific evidence.

Reading Your Test Report: What Information Should Be Included

A complete paternity test report should contain specific information that allows you to understand the testing process and verify the results. Knowing what to look for helps you evaluate whether the testing was done properly and whether the conclusions are supported by the data shown.

Your report should identify all parties tested by name and/or identification number. It should clearly state which genetic locations (loci) were tested—this number typically ranges from 16 to 25 for standard paternity tests. The report should show the alleles found at each location for the child, the mother (if tested), and the tested man. This allows you or someone else to verify the lab's interpretation by checking the inheritance pattern yourself.

Look for the paternity index or probability calculation section. This should explain the statistical method used and show how the final percentage or ratio was derived. A quality report explains the result in plain language, such as "Based on the genetic markers tested, [Name] cannot be excluded as the biological father" or "[Name] is excluded as the biological father based on genetic markers at [specific locations]."

The report should include information about the testing lab, its accreditation status, and the credentials of the individuals who performed and interpreted the testing. Many reputable labs display accreditation from organizations like the American Association of Blood Banks (AABB) or other recognized certifying bodies. The date of testing, sample collection, and report generation should all be clearly stated.

Check whether the report notes any limitations or issues with the samples. If samples were degraded, if results are inconclusive, or if retesting is recommended, this should be explicitly stated with explanation. Quality labs are transparent about any technical issues encountered.

Practical takeaway: Before accepting results, verify that your report includes tested locations, individual allele readings, calculation methodology, and lab accreditation information. If your report lacks any of these elements, contact the lab to request a more complete version.

What Can Affect Test Accuracy and When Results May Need Verification

While DNA paternity tests are highly accurate when performed properly, several factors can affect the reliability of results. Understanding these factors helps you recognize when retesting might be needed or when results should be interpreted with caution.
